THE ISOLATION,PURIFICATION AND CHARACTERIZATION OF AN EXTRACELLULAR PROTEOGLYCAN FROM ISARIA FARINOSA

Abstract:
An extracellular proteoglycan PG was isolated from the culture filtrate of Isaria farinosa (Dicks) Fr. The crude preparation was obtained by ultrafiltration, three volumes of ethanol precipitation and acetone drying, and purified by resin 122#(H), Amberlite resin IR-45(OH) and Sephadex G-150 chromatography. PH was homogenous, as determinedly gel filtration on Sepharose 4B, acetic fibre paper electrophoresis and polyacrylamide gel electrophoresis. The average molecular weight of PG was 111000. PG is mainly consisted of polysaccharides (92.35%) together with small proportion of proteins (7.61%). PG was investigated by G.L.C. and I. R. spectroscopy. The results indicated that PG was composed of D-mannose and D-galactose in the molar ratio of 5.93:1 and linked mainly by a -glycoside bond. PG shows inhibitory activity against implanted Sarcoma 180 in mice. The percentage of inhibition is 22.2%.
