STUDIES ON CYCLOSPORIN 8562

Abstract:
Cyclosporin 8562 was produced by Tolypocladium sinense C. L. Li strain 8562 which was isolated from fresh material Cordyceps sinensis (Berk.) Sacc, collected from Diqing Tibetan Autonomopus Prefecture, Yunnan province. The cultures of Tolypocladium sinense C. L. Li strain 8562 was extracted with an equal volume of acetic ether and concentrated to dry under reduced pressure. The light yellow crude samples was separated by silica gel column chromatography using acetic ether water saturation as the eluent. The crude compound was separated into two components: 8562-component 1 and 8562-component 2. Each component was then further purified by sephadex LH-20 chromatography using methanol as the eluent. They are as colourless amorphous power. Purified sample 8562-component 1 and 8662-co-mponent 2 were identified as Cyclosporin A and cyclosporin C respectively by TLC, UV, IR, HPLC, FABMS and 1H-NMR. It could be concluded that the 8562-component 1 and 8562-component 2 of the compound produced by Tolypocladium sinense was identical with cyclosporin A and cyclosporin C. The antiobiosis of 8562-component 1 was investgated and compared with that of known cyclosporin A. The 8562-component 1 was shown in this test to have a obvious antifungal activity against some imperfect fungi and yeasts.
