Screening, identification and properties study of marine bacteria producing alkaline cellulase

Abstract:
Using CMC plate screening method,a marine strain QM11 that can produce alkaline cellulase is isolated from the seawater of Qingdao coast.According to the 16S rDNA analysis,the strain belongs to the Cytophaga fucicola.Studies of its biological characteristics have shown that the optimal growth temperature is 27 ℃,and the growth temperature range is between 4℃and 48℃,which indicates it iS a cold-resistant bacterium.Media of pH7.0~8.0 and 3.0%NaC1 are the most suitable for the growth and enzyme production.The optimal temperature for QM11 producing alkaline cellulase is 40℃.The best reacting pH is 9.0.and the strain has a high enzyme activity and a good stability in the basic conditions.Studies also show that Mn2+ and Fe3+ could stimulate the enzyme reaction.while Cu2+ and Pb2+ inhibit the reaction.
