STUDIES ON KARYOTYPE AND C-BANDED, G-BANDED CHROMOSOMES OF Haemolaelaps casalis

Abstract:
This paper first presents the studies on karyotype and C-banding, G-banding of the chromosome in Haemolaelaps casalis. An analysis of the karyotype of H.casalis was carried out on the basis of the chromosomes of eggs by cellophane squash technique, taking eggs by caesarean section, using Giemsa stain. The results show that the karyotype of H. casalis is haplodiploidy, i. e., haploid chromosome number is 7 (n = 7), while that of diploid chromosome is 14 (2n=14). C-banding analysis of egg cellophane squashes of H. casalis by barium hydroxide-Giemsa banding technique (CBG) showed that constant C-bands would appear at several positions such as No. 1, 2, 4 and 5 chromosomes, but nonconstant C-bands would do at No.3, 6 and 7 chromosomes. According to the result of C-banding pattern, of these chromosomes of H. casalis, No. 1, 2 and 7 were submetacentric (sm), No. 4 was subtelocentric (st), No. 6 terminal region centric (t) and No. 3 and 5 were terminal point centric (T). The G- banded chromosomes of H. casalis by trypsin- Giemsa banding technique (GTG) evidenced that the position were C-bands have appeared reveal dark bands, with chromosome arms ranging from 2 to 10 dark bands.
