THE ESTABLISHMENT OF THE BLACK MUNTJAC (MUNTIACUS CRINIFRONS) CELL LINE AND ITS GROWTH AND CYTOGENETIC CHARACTERS

Abstract:
Black muntjac is only found in China. Its chromosomes are particularly large and the diploid number is 2n = 8 , 9 . There are three pairs of chromosomes similar in size and mophology. We have established a fibroblast diploid cell line from the female fetal lung tissue. These cells were through 149 passage over one year. The growth rates and mitotic index were examined. The formation time of confluent monolayer by various cell densities was also studied. The results indicated that cells of passage 86 grew much better than those of passage 10. The patterns of G-, C- and Ag-bands have also been analysed. From the passage 90, we found a new subpopulation of cell with the karyotype (2n= 8 ) contaning two isochromosomes of different sizes. The new subpopulation has been isolated. The G-banding analysis demonstrates that the two isochromosomes with different sizes are derived from the long arm and the short arm of No 1 chromosomes.
