ANALYSIS OF PolyU·polyA·polyU STRUCTURE BY RAMAN SPECTROSCOPY AND LATTICE DYNAMICS

Abstract:
In recent years,increasing interest has been focused on triple helical nucleic acids due to their importance in therapeutic,diagnostic,and biotechnological applications.In this paper,a Raman spectrum of polyU·polyA·polyU which is one of the most important nucleic acid was presented with high signal noise ratio.A normal mode analysis of A-conformation structure model of polyU·polyA·polyU proposed by Arnott basing X-ray diffraction was carried out by the lattice dynamics,and each Raman band was assigned by potential energy distribution(PED).The result shows each chain of polyU·polyA·polyU possesses similar vibrational frequencies and PED.No Raman band belongs to only one chain.The fact that the calculated frequencies are good agreement with the observed Raman spectrum indicates that polyU·polyA·polyU in solution probably possesses A-conformation structure model which was represented by Arnott.
