FITTING CALCULATION OF FLUORESCENCE RESONANCE ENERGY TRANSFER EFFICIENCY BY MEANS OF EMISSION SPECTRA OF DONOR-ACCEPTOR PAIR

Abstract:
Fluorescence resonance energy transfer (FERT) is widely used in studies of biomolecular structure and dynamics.We present a simple method to calculate FRET efficiency by using emission spectra of the donor,the acceptor and the donor-acceptor pair.This method holds provided that the donor is selectively excited and the concentration ratio of the acceptor and the donor is known.A GFP based donor acceptor pair was taken as an example to demonstrate the methods.The obtained FRET efficiencies of Cameleon at zero calcium and saturating calcium are consistent with those obtained by other methods.
