Quantitation of Rubisco activase by single radial immunodiffusion.

The antibody against Rubisco activase was produced with rabbit after purification of the enzyme and the technology of single radial immunodiffusion for the enzyme was set up. The contents in the range of 0 to 200 μg/mL of the enzyme are proportional to immunodiffusion precipitation zone areas. The method has been used for determining the content of Rubisco activase in rice leaf when the leaf extract was concentrated with 35% saturated ammonia sulfate. Rubisco activase contents in rice flag leaf decreased slowly during the first nine days and thereafter declined sharply after heading.
