A STUDY ON GENE TRANSFORMATION OF IMMATURE WHEAT EMBRYOS BY ELECTROPORATION

Abstract:
By use of the HPES-3 pulse discharging electroporation system that is developed by our laboratory, we have transferred the bar gene(phosphinothricin acetyltransferase gene) in plasmid pDM302 into the immature embryos of a spring wheat cultivar (Triticum aesivum L. ) "T2003". After selection for 2~ 3 months, we have obtained the ppt (phosphinothricin)-resistant callus, from which regenerated a wheat plantlet. PCR analysis and DNA dot hybridization showed the foreign bar gene had been integrated into the genomes of the transformed wheat plantlet.
