A New Method for Callose Fluorescence Observation of Microspore Mother Cells During Meiosis

Abstract:
Staining with the improved carbol fuchsin-aniline blue fluorescence method is a convenient and efficient way to observe the behavior of the callose wall of microspore mother cells during meiosis. The photos are vivid and colorful and showred cytoplasm, carmine chromosomes, and kelly callose; the contrast is strong and demonstrates a 3-D effect. Cytoplasm and chromosomes appear as red fluores cenceunder the excitation of blue light, and the chromosomes are clear on staining with only improved carbol fuchsin. Therefore, improved carbol fuchsin can be used as the fluorochrome to nuclear DNA to reduce the costs of the use of DNA-specific fluorochrome such as DAPI and H33258.
