THE DYNAMIC CHARACTER OF INTRODUCTION OF DNA INTO CELLS BY ELECTROPORATION

Abstract:
In this study E. coli HB101 is used as the cell model. It has been investigated to causecell electroporation with RC electric pulses of different strength and different width andintroduce various kinds of plasmids into cells. These plasmids contain the long thready closedcircular plasmids of pUC-18, pCP10,which are 422nm and 1680nm long respectively, and theplasmid of pCP10 condensed with spermine, which is a kind of toroidal particle with the diameterof 88nm, Their transformation efficiencies have been mensured respectively. The experimentalresults manifest the dynamic character of introduction of DNA into cells by electroporation.
