Effeet of DS-1226 on Myelosuppression of Mice Caused by Cyclophosphamide

Abstract:
This study aimed to investigate the effect of capsule formulation of DS-1226 on cyclophosphamide-induced myelosuppression in mice. Mice were randomized into five groups: the control group, the CTX group and three treatment groups respectively at the dose of 37.5, 75, and 150 mg·kg-1·d-1 DS-1226 capsule content. After one bolus intraperitoneal injection of 200 mg·kg-1 cyclophosphamide, the treatment groups were orally administrated with DS-1226 capsule decoction at different dosages for 8 days, while the control group and the CTX group were given distilled water. The general condition, peripheral blood cell counts, bone marrow nucleated cells, bone marrow structure, the thymus index and the lymphocyte transformation rate were examined. The results showed that DS-1226 could improve the general condition, increase peripheral blood counts of leucocytes, lymphocytes, erythrocytes and bone marrow nucleated cells, promote hematopoiesis in bone marrow cavities, and stimulate the thymus index and the lymphocyte transformation rate. The hematopoietic effect of DS-1226 may help mitigate cyclophosphamide-induced myelosuppression and promote immunity in mice.
