Optimization of Extraction Process of Polysaccharide from Inonotus obliquus by Uniform Design

Abstract:
This study aimed to optimize the extraction techniques of polysaccharide from Inonotus obliquus by uniform design. The content of I. obliquus polysaccharides was determined by spectrometry method, which takes anhydrous glucose as standard and 5% phenol and sulfuric acid as color-developing agent, in order to investigate the extraction and alcohol-purification technologies of I. obliquus polysaccharides. The extraction and alcohol-purification technologies of I. obliquus polysaccharides were optimized by employing the uniform design method. Results showed that the best extraction technology was when the I. obliquus and water ratio is 1:15. And the extraction should be given twice and 90 minutes for each time. The best alcohol-purification technology was to add alcohol into the concentration of 1.0 g/mL crude herb to 65% with vigorously rabbling. The conclusion is that the extraction and alcohol-purification technologies of I. obliquus polysaccharides optimized by uniform design method are economical, stable and efficient.
