Separation of Phillyrin from Fructus Forsythiae by High-Speed Counter-Current Chromatography

Abstract:
Phillyrin,an active component in the Fructus Forsythiae,was successfully separated from Chinese herbal medicine Fructus Forsythiae by High-Speed Counter-Current Chromatography(HSCCC).The solvent system used for separation was consisted of carbon tetrachloride-chloroform-methanol-water.The upper phase was used as stationary phase and the lower phase was used as mobile phase.The apparatus was rotated at 900 rpm and the mobile phase of solvent was pumped through the column at a flow rate of 1.2 mL/min.The identification by ESI-MS and 1H-NMR proved that the component was phillyrin,and the purity was 98.8%.It is concluded that HSCCC can be used in the separation of phillyrin.
