Study on the Optimized Purification Process with Macroporous Resin Technology for Total Timosaponin of Radix Polygalae

Abstract:
This study was aimed to optimize the purification process with macroporous resin technology for total timosaponin in Radix Polygalae. The total timosaponin was taken as characteristic marker. The process of alcohol concentration, alcohol consumption and extraction time were optimized by using orthogonal design. Then, the effect on purification result was investigated on aspects of the concentration of sample, adsorption velocity, volume of sample, cleaning solvent, concentration of elution and volume of elution on the selected type of macroporous resin. The result showed that extraction with 50% ethanol 12 times as much as the total timosaponin for 2.0 h and twice was more effective. Extracting solution was put through AB-8 macroporous resin column (diameter-to-length ratio is 1:8), at the adsorption velocity of 1mL/min. Then, 6 times the volume of caustic soda solution with the concentration 0.2mol/L was used to wash, and it was washed again with distilled water to neutral effluent. The elution used 3 times the volume of 80% ethanol. It is concluded that this process is stable and practicable. The macroporous resin technology can be used in the extraction and purification of total timosaponin in Radix Polygalae.
