Label-Free Detection of Artemisinin and Aristolochic Acid Based on Microcantilever Sensor

Abstract:
A novel technology for label-free detection of Artemisinin and Aristolochic acid antigen-antibody specificity binding based on microcantilever sensor platform is reported in this paper. Firstly, Sulfhydryl Artemisinin /Aristolochic acid antibody was immobilized on gold-coated side of a microcantilever by self-assembled monolayer technique. Then, the deflections of microcantilever corresponding to different levels of concentrations of Artemisinin /Aristolochic acid antigen could be real-time monitored by using optical lever technique. As a result, the sensitivity of detecting Artemisinin and Aristolochic acid is 100ng/ml and 500ng/ml, respectively, which is corresponding to the tendency of their Standard inhibition curves obtained in ELISA test. The sensitivity of detecting Artemisinin is higher due to its antibody is more active than that of Aristolochic acid antibody. Results show that it is feasible for detection of Artemisinin and Aristolochic acid by microcantilever sensor platform.
