HYPOCRELLIN-B (HB) SENSITIZED PHOTODAMAGE TO MURINE ASCITIC HEPATOMA CELLS

Abstract:
In this paper, we primarily studied the HB-sensitized cytotoxicity to murine ascitic hepatoma cell (AH). The results showed that the cell mortality was roughly proportional to the square root of HB concentration. The ability to kill AH cell among hypocrellin A (HA) , HB, protoporphyrin (PP) , and he-motoporphyrin (HP) are in the following order: PP>HB>HA>HP. The ultr-astructure observation using TEM showed that the plasma membranes, mit-ochodria and endoplasmic recticulus were probably the main targets of photose-nsitization. After 10 minutes irradiation in the presence of HB, obvious ves-iculation and dramatic rupture of plasmic membranes were observed, but the nucleic membranes and nuclei did not change significantly. SEM revealed that the shape of the photodamaged cells changed obviously and the microvilli on the cells disappeared. By using active oxygen quenchers, spin trapping and spin silence techniques, we proved that singlet oxygens, superoxide anion radicals and hydroxyl radicals were involved in the process of HB-photosensitized cytotoxi-city.
