ELECTROFUSION BETWEEN PROTOPLASTS OF CABBAGE AND CHINESE CABBAGE

Abstract:
Electrofusion was performed between mesophyll protoplasts of cabbage (Brassica oleracea var. capitata, with Raphanus 'Ogura' cytoplasmic male sterility) and suspension protoplasts of Chinese cabbage(B. Campestris var. pekinensis). With a wide-interval(1.5 mm between two electrodes) electrofusion chamber,the operation is simple and the highest ratio of heterofusion up to 46±6% could be obtained, among which the percentage of di-heterofusion was 11±1%.When heterokaryons were cultured with unfused protoplasts at a density of 104/ml,about ten percent of heterokaryons resulted in first division within 24 h. Division ratio of heterokaryons reached 70% after 48 h in culture. Heterokaryons having chloroplasts from mesophyll of cabbage and cytoplasm from suspension of Chinese cabbage were isolated manually by dilution of the culture at the first or second day. Some of them undergone sustained division at a density as low as 102/ml or even in a single heterokaryon culture.The calli derived from such a way regenerated roots on a differentiation medium.
