STUDIES ON THE IRREVERSIBLE INHIBITION OF CREATINE KINASE FROM HUMAN MUSCLE BY DTNB

Abstract:
The irreversible inhibition of creatine kinase from human muscle during modification with DTNB has been studied. It has been found that four SH groups per enzyme molecule, which is different from rabbit muscle CK, could be modified. The result handled by Tsou's plot suggests that one of the four reactive SH groups is fast reactive and nonessential for activity, two of them are slow and essential, and another is very slow and nonessential. A series of kinetic constants have been measured, with the method of kinetics of irreversible modification of enzyme activity in the presence of inhibitor put forward by Tsou, and the acting mechanism of DTNB has also been discussed.
