Injury mechanism of primary cultured hippocampus neuron by electromagnetic radiation

Abstract:
To investigate the injury mechanism of primary cultured hippocampus neuron exposed to X-band high power microwave,S-band high power microwave and electromagnetic pulse radiation,primary hippocampus neurons were cultured in vitro,and cell model of electromagnetic radiation was established.Annexin V-PI double labeling and flow cytometry were applied to detect neurons apoptosis and death;Atomic force microscope was used to detect the surface appearance of cellular membrane;Fluo-3-AM fluorescent probe load and laser scanning confocal microscopy were employed to measure Ca2 ]i in neurons.The results indicated that,after radiation,the apoptosis and death rate of hippocampus neurons both increased,and the death rate increased more significantly;The surface roughness and perforation of cellular membrane increased;Ca2 ]i in neurons increased significantly as well.All the changes above showed an attenuating trend in the radiation groups as X-HPM?S-HPM?EMP.It was suggested that perforation increased membrane permeability,and then increased the inflow of extracellular Ca2 ,even resulted in intracellular calcium overload.All these may play an important role in the mechanism of hippocampus neuron apoptosis and death induced by radiation.The injury degree of hippocampus neurons by the three kind of electromagnetic radiation showed a direct correlation with radiation frequency.
