INCORPORATION OF EXOGNOUS LAMBDA DNA INTO ASSEMBLED NUCLEI IN XENOPUS CELL-FREE SYSTEM

Abstract:
By adding exogenous Lambda DNA to a cell-free system prepared form Xenopus eggs, nuclei were assembled.the intra-nuclei and extra-nuclei DNA were isolated at different Lambda DNa;and the contentof intra-nuclei and extra-nuclei DNA were increasing and decreasing, respectively, during the course of assembly. The extra-DNA was sensitive to DNase I, but intra-DNA could not be digested due to the protection of nuclear membrane. The restriction pattern of intra-DNA was the same as that of Lambda DNA. We conclude that the exogenous Lambda DNA is incorporated into the assembled nuclei.
