CULTIVATION OF VASCULAR SMOOTH MUSCLE CELLS ON CHITOSAN AND GELATIN FILMS

Abstract:
Chit os an, a positively charged polysaccharide, has shown to be a structural material o f promise for a number of tissue engineering applications. The role of the degr ee of deacetylation (DD) on some biological properties of chitosan films was in vestigated in vitro. The potential of chitosan, gelatin, and gelatin-chitosan c omplex materials for controlling the proliferation of vascular smooth muscle cel ls (VSMC) was also evaluated. Material films were seeded with VSMC for culture up to 7 d. In 1 and 5 d culture, phase contrast microscopic images of the cells on each surface were digitally captured. Cell growth was evaluated by the MTT a ssay at 24?72 and 120 h. The results indicate that all materials were biocompat ible with VSMC and VSMC proliferation increased when the DD of chitosan films i ncreased. In conclusion, DD plays a profound role in VSMC cell adhesion and pro liferation.
