Selection of Resistant Mutants for Helminthosporium maydis via Tissue Culture in Maize

Abstract:
Embryogenic calli derived from the commercial selfline Huangzhaosi of maize were transferred to medium supplemented with toxin from Helminthosporium maydis for the selection of resistant cells. After three selections, toxin resistant calli were produced. The toxin showed obviously effects on the proliferation and differentiation of callus cells. On the medium added the toxin, plantlets were regenerated from toxin resistant calli. The progenies of one regenerated plant showed significantly stronger resistance than Huanghzaosi, It was concluded that the toxin resistant character could be stably inherited by the analyses of data from backross plants.
