EFFECT OF ACLACINOMYCIN (ACM) ON CELL CYCLE OF L_(1210) LEUKEMIA CELLS BY DUALPA-RAMETER FLOW CYTOMETER

Abstract:
The effect of ACM A and ACM B on cell cycle progression and RNA contents of L1210 cells were compared in vivo.By using dual parameters flow cytometer to measure cellular DNA and RNA contents simultaneously, early and middle S cells were accumulated 8 hr after treatment with 1/2,5 or 1/4.0 LD50 of ACM A or ACM B, this showedthat the DNA synthesis rate was decreased in the early and mid S phase cells. The DNA synthesis inhibition was reversible, so S cellls entered to G2 phase and caused accumulation of G2 cells due to the progression of G2 to M was blocked or slowed down by 24 hr treatment. At the same time, the RNA content of cells blocked in G2 phase were increased predominately and caused the positive unbl-ance growth. In contrast, the RNA content of S phase cell was not increased concomitantly with the increment of DNA content and caused the negative unbalance growth.In cluclusion, the effect of ACM A and ACN B on the cell cycle progression of L1210 cells in vitro or in vivo is almost the same, whereas the inhibition of RNA synthesis of L1210 cell in vivo are recovered in G2 and G1 cells but not in the S phase cells treated with ACM A or B by 24 hr.
