Animal Cell Death Rates in Sparged Bioreactors
|
Abstract:
The experimental results obtained from this work demonstrated that the cell death rate in sparged bioreactors is proportional to the gas sparging rate and inversely proportional to the bioreactor volume. It was found that the bubble size has two effects on cell death rate. The one is to affect the bubble surface area production rate and the other is to influence the adsorptivity of the cells on bubble surface. The optimum bubble diameter at which the cell death rate reaches minimum is about 5mm. The effect of media formulations on cell damage was also investigated. Both the serum and Pluronic F68 were found to reduce the cell death rate significantly. And as the concentration of Pluronic F68 increases to 0. 1% , the value of kd drops to zero. All the experimental data are in excellent agreement with the bioreactor design model developed in the previous paper.
