QUANTITATIVE ANALYSIS OF THE PRIMARY STRUCTURE-FUNCTION RELATION OF CARDIOTOXIN FROM COBRA VENOM

Abstract:
A quantitative analysis method for the determination of primary protein structure and function relation was proposed. The biological activity of variant acid residues at the identity side and different sites of the peptide chain and the location of the active region and sites can be estimated quantitatively by the establishment of structure-function equation and the identfication of replacement parameters and sites parameters. The relationship between the primary structure and 50% mice lethal dosage of 24 samples of cardiotoxin from cobra venom as well as the relationship between the primary structure and the 63% depolarization dosage of 19 samples of cardiotoxin for cultured rat myoball were analysed. The result suggested that the site 4,10,26,29 and 60 were of great importance to the biological activity and the site 10 and 29 at the 1st and the 2nd loops may be involved in the target combination.
