A STUDY ON THE ANTICANCEROUS CHARACTERISTICS OF THE TRAGETED LIPOSOMES CONTAINING LINOLEIC PLATINUM

Linoleic platinum-containing untargeted and targetal liposomes learing antihuman Breast cancer cell monoclonal antibody Me Ah Gp-1D8 on their surface are prepared by sonication,The tissue distribution of linoleic platinum encapsulated liposomes and its efficacy of tumor inhibit after injection i.p. tumor-boring nude mice as compared with that of free linoleic platinum were investigated. The result indicated that the content of linoleic platinum of targeted liposomes in the tumor thenes was apparently higher than that of free linoleic platinum graup.However, the conent of the former was lower than the latter in heart, liver, lung, spleen and kidney. Free linoleic platinum or targeted liposomes were injected i.p. into nude mice (4mg/Kg body weight) at day 6,12,and 24, respectively, and the data were obtained at 40 day after inoculation of breast cell. The result shows that linoleic platinum targeted liposomes has advantage over free linoleic platinum in some respects such as the amount of survived animals, the ralio of tumor occurrence and the rate of tumor growth. The interaction of linoleic platinum targeted liposome with tumor cell membranes has been investigated by mothod of MSL spin Labeling. In the presence of linoleic platinum the ratio W/S of breast cancer cell membranes and S180 Cell membranes labeled with MLS were changed. The result indicates that linoleic platinum can react with mercapto groups and change the conformations of membranes proteins of tumor cells.
