SCANNING TUNNELING MICROSCOPY IMAGING OF DMPC MONOLAYER AND CYTOCHROME C OXIDASE RECONSTITUTED IN LIPID BILAYER

Abstract:
We have employed a SSX-1 STM to determine the structural properties of a) DMPC monolayer supported on graphite in native environment and b) CL/DPPC liposomes reconstituted with cytochrome c oxidase, coating with a thin Au film afetr air-drying. Images of DMPC monolayer show long range positional as well as or-ientational order and individual headgroups are resolved. Images of liposomes re constituted with cyt. c oxidase show the enzyme molecules exist in membrane in "dimer" and the fine structure of enzyme are also resolved. The results indicate that STM has a great potential in membrane research.
