Abstract:

at the moment the chondrocytes culture have a wide clinical application in the treatment of pathologies of the articular cartilage,as well as in the reconstruction of auricular and nasal cartilage using natural or synthetic substrata. in view of the importance of this culture type, the following work had as purpose to standardize the methodology for the establishment of the cultures of human and chicken chondrocytes, evaluating the growth of these cells in culture in monocapa and in three-dimensional matrix of collagen type i, by means of histochemistry, inmunohistochemistry and bio- chemistry technical. the human and chicken chondrocytes cultivated on plastic to high densities they showed a growth in monocapa,standing out a cellular appearance of polyhedrical type with prominent spaces among them,where were de- tected components of the matrix extracelular such as glicosaminoglycans (gag),but this production is low and most of these diffuse to culture medium due to the lack of a matrix that retains them.on the other hand,the matrix of collagen type i, offer to the human and chicken chondrocytes a three-dimensional space that simulates the "in vitro" conditions,and for that reason the chondrocytes maintains its spherical morphology and they synthesize characteristic matrix components such as sulfated gag and collagen type ii that are markers of the stability phenotypical of these cells.
