New methods of removing debris and highthroughput counting of cyst nematode eggs extracted from field soil

Upender Kalwa, Christopher M. Legner, Elizabeth Wlezien, Gregory Tylka, and Santosh Pandey
The soybean cyst nematode (SCN), Heterodera glycines, is the most damaging pathogen of soybeans in the United States. To assess the severity of nematode infestations in the field, SCN egg population densities are determined. Cysts (dead females) of the nematode must be extracted from soil samples and then ground to extract the eggs within. Sucrose centrifugation commonly is used to separate debris from suspensions of extracted nematode eggs. We present a method using OptiPrep as a density gradie...

Scan4CFU: Low-cost, open-source bacterial colony tracking over large areas and extended incubation times

Santosh Pandey, Yunsoo Park, Ankita Ankita, Gregory J. Phillips
A hallmark of bacterial populations cultured in vitro is their homogeneity of growth, where the majority of cells display identical growth rate, cell size and content. Recent insights, however, have revealed that even cells growing in exponential growth phase can be heterogeneous with respect to variables typically used to measure cell growth. Bacterial heterogeneity has important implications for how bacteria respond to environmental stresses, such as antibiotics. The phenomenon of antimicrobia...

Morphologycal and Molecular Partial Histone-H3 Characterization of Bintan Sea Snail Gonggong (Strombus sp.) as a Species Validation

Lily Viruly, Nuri Andarwulan, Maggy T. Suhartono, Mala Nurilmala
Sea snail gonggong is an icon of Tanjungpinang-Riau Islands Province. It is a favorite seafood item in Riau Islands Province, and is high economic value but not known widely yet. Until now, sea snail gonggong has been highly exploited but the research on this snail is very limited. The aim of this study was to morphology and molecular characterization of Bintan gonggong snail (Strombus sp.) as a species validation. Bintan gonggong snail included thick-shelled gonggong and thin-shelled gonggong. ...

Molecular Characterization and Phylogenetic Relationships of Melon (Cucumis melo L. Meloni Cultivar) Based on Expression of Andromonoecious Gene

Eka Wasi’ Al-Mughni, Budi Setiadi Daryono
Melon (Cucumis melo L.) is one of horticultural commodities with good economic prospect for melon farmers because of several advantages. 'Meloni' is result of crossing between ‘Sun Lady-3’ and ‘PI 371795’ by Genetics and Breeding Laboratory, Faculty of Biology, UGM. The 'Meloni' plant produces females flowers that expressed by CmACS-7 gene and it can be expected that melon plants 'Meloni' included in the primitive cultivars, while 'Meloni' is a cultivation cultivar. The aim of this research is t...

Anatomical Properties and Near Infrared Spectra Characteristics of Four Shorea Species from Indonesia

Danang Sudarwoko Adi, Sung-Wook Hwang, Dwi Ajias Pramasari, Yusup Amin, Hairi Cipta, Ratih Damayanti, Wahyu Dwianto, Junji Sugiyama
This study investigated the anatomical properties and absorbance characteristics of NIR spectra of four Shorea species from Indonesia. Macroscopic section revealed that Balau has similarity with Heavy Red Meranti, whereas White Meranti was almost identical with Light Red Meranti. All of the woods have diffuse porous and axial resin canals in tangential lines at the microscopic level. Original NIR spectra of Shorea species showed different absorbance characteristic. Wood density was assumed to be...
