Protoporphyrin IX is a dual inhibitor of p53/MDM2 and p53/MDM4 interactions and induces apoptosis in B-cell chronic lymphocytic leukemia cellsDOI: 10.1038/s41420-019-0157-7 Abstract: a F2H?-analysis of disruption of interactions of p53/MDM2 and p53/MDM4 by PpIX and nutlin in U2OS cells, when MDM2 or MDM4 is tethered in the nucleus. U2OS cells were co-transfected with LacI-GFP-MDM2 or MDM4 and RFP-p53 for 8?h and then treated with PpIX or nutlin for 16?h. Control interaction values are normalized to 100% and are shown as a red line. Averaged interaction values for the treated cells are plotted for p53/MDM2 interaction (left, green bars) and p53/MDM4 interaction (right, blue bars). Data represented as mean?±?s.e.m., n?=?5–6, *p?<?0.05, **p?<?0.01, ***p?<?0.001, Student’s t-test. b Rescue of the p53 transcription activity in the presence of MDM4 by PpIX but not nutlin as assessed in the yeast-based reporter assay. The average light units relative to the transactivation activity of p53 alone and the standard errors of at least five biological repeats are presented. The Student’s t-test was performed for statistical analysis with p?≤?0.05. c PpIX (1?μg/ml) disrupts p53/MDM4 interactions in HCT 116 colon cancer cells as assessed by immunoprecipitation. The data is a representative of two independent experiments. Cropped line represents the site were the membranes were cut. X-sample not relevant to this stud
