灵芝多糖对T24荷瘤裸鼠化疗效果及其免疫逃逸的影响

Abstract:

摘要 目的 探讨灵芝多糖(ganoderma lucidum polysaccharide, GLP)对膀胱癌化疗效果的影响及 其机制。 方法 建立T24荷瘤裸鼠模型,分组给药后测定脾脏指数,利用流式细胞仪检测小鼠脾脏和 血液中相关淋巴细胞亚群所占比例,采用双抗体夹心(ELISA)法检测荷瘤小鼠血清IL-10、VEGF、 TGF-β、IL-2和TNF-α含量,应用反转录-聚合酶链反应(RT-PCR)和Western blot检测小鼠脾脏IL-2 和TNF-α的表达。 结果 在荷瘤裸鼠模型中,灵芝多糖(200 mg/kg)灌胃18 d后可显著增加脾脏指 数(P<0.001);顺铂+灵芝多糖组中小鼠脾脏及血液中的淋巴细胞(CD4、CD8、NK及DC)都较 顺铂组提高,而髓源性抑制细胞(myeloid derived suppressor cell, MDSC)含量降低。通过ELISA检测 发现灵芝多糖能明显增加外周血液中相关免疫因子(IL-10、VEGF、TGF-β、IL-2和TNF-α)的含量 (P<0.05)。 RT-PCR及Western blot的结果也证实了在灵芝多糖可增强小鼠化疗后脾脏中IL-2和TNF-α 的表达。结论 灵芝多糖能通过提升脾指数,增加脾脏和外周血相关淋巴细胞及免疫因子的含量,并 通过增强血清中IL-2和TNF-α的表达来提高小鼠的免疫力,增强抗肿瘤作用,其机制可能与灵芝多糖 降低免疫逃逸功能相关
