七叶鬼灯檠根中总多酚的提取及抗氧化活性的研究

Abstract:
以七叶鬼灯檠干燥根为原料,比较不同提取方法、不同溶剂对总多酚提取量的影响。并采用L9(34)正交试验对七叶鬼灯檠根中总多酚的提取工艺参数进行优化,得出回流提取法,以乙醇为提取取溶剂下提取量较高。在单因素试验结果上考察不同因素对七叶鬼灯檠根中总多酚提取量影响大小须序为提取时间>提取温度>乙醇浓度>液料比;最佳提取工艺条件为提取时间90 min、提取温度90℃、乙醇60%、液料比50∶1,提取4次,总多酚的提取量为123.599 mg/g。用FRAP法对多酚粗提取物的抗氧化活性进行测定,总多酚提取物以总多酚计的抗氧化活性为14.076 mmol FeSO4/g总多酚,相当于1.079 g Vc的抗氧化活性。
Technological process of extracting polyphenols from the roots of Rodgersia aesculifolia was examined by single factor experiment and L9(34) orthogonal design. It was found that high yields of polyphenols would be achieved by using ethanol as solvent and refluxing. The influences of the relative factors were in the order of time> temperature > ethanol concentration > material to solvent ratio. The optimum extraction conditions were determined as time 90min, 90℃, 60% ethanol, the ratio of material to solvent 1∶50, 4 times. Under these conditions, the total polyphenols extraction the quantity was the 123.599 mg/g (dry roots). The antioxidant activity of crude extracts was measured with FRAP method and the result showed that the polyphenol antioxidant activity is 14.076 mmol FeSO4/g polyphenols accounted by total polyphenols and was equivalent to the antioxidant activity of 1.079g Vc
