Abstract:

为获得可应用于农田土壤重金属污染治理的铅(Pb)降解功能菌株,从陕西商洛铅锌矿区周边农田土壤中分离筛选出1株抗铅细菌,命名为PS8,对其形态特征、生理生化、16S rDNA同源性及抗铅性能进行研究。结果表明,菌株PS8为革兰氏阴性细菌,菌落呈圆形、淡黄色;最适生长温度为30 ℃,最适pH为7.0;可还原亚硝酸盐,氧化酶、接触酶反应呈阳性;与粪产碱菌Alcaligenes faecali高度同源;在Pb2+浓度为0.5和1.0 mmol/L时,去除率分别为99.08% 和97.24%。菌株PS8对重金属铅具有高抗性和较强降解能力,是高效、环境友好的土壤重金属铅污染微生物修复菌株。
In order to obtain resistant bacteria for removing heavy metal in polluted cultivated field, a lead-resistant strain, named PS8, was isolated from metal-polluted soil in a cultivated field near lead-zinc mining area in Shangluo, Shaanxi province. Based on 16S rDNA sequence analysis, PS8 was highly homologous to Alcaligenes faecali by studying the morphological, physiological-biochemical properties, the results showed that PS8 was gram negative, circle-shaped, yellow color and its most suitable growth condition was 30 ℃ pH 7.0. In addition, Nitrite reduction test, oxidase test and catalase test were all positive. The Pb2+ removal effect of PS8 was 99.08% and 97.24% respectively in 0.5 mmol/L and 1.0 mmol/L lead concentrations. In conclusion, PS8 is an effective, environmental- friendly strain and have remarkable value to remediate heavy metal contaminated soil.
