西地那非对西藏小型猪慢性肺动脉高压模型体循环压力的影响*

Abstract:
目的:观察西地那非对西藏小型猪慢性肺动脉高压(PH)模型体循环压力的影响。方法:18头2月龄西藏小型猪随机分为正常对照组、模型组和治疗组,每组6头。模型组和治疗组小型猪腹腔注射野百合碱42 d用以制备PH模型,正常对照组给予相同剂量溶剂。第42天,超声心动图扫查估算肺动脉压。第43天开始,治疗组喂食西地那非(6.0 mg?kg-1?d-1),共21 d。第64天超声心动图扫查估算动物肺动脉压,左颈动脉插管测定体循环压力。结果:与对照组比较,模型组肺动脉压和体循环压力均明显升高(P<0.05); 与模型组相比,治疗组肺动脉压均降低,同时体循环压力亦明显下降,甚至低于正常对照组(P<0.05)。结论:西地那非在降低肺动脉压的同时也降低体循环压力。
Aim: To investigate the effects of sildenafil on the systemic blood pressure of Tibet minipig models with chronic pulmonary hypertension(PH).Methods: A total of 18 Tibet minipigs were randomly allocated into control group, model group and treatment group, 6 in each group. The Tibet minipigs in model group and treatment group were treated with monocrotaline(MCT)for 42 d to establish PH model.Since day 43, the treatment group was treated with sidenafil(6.0 mg?kg-1?d-1)for 3 weeks. Conventional transthoracic echocardiography was performed at day 42 and day 64.The systemic blood pressure was measured by left carotid artery cannulation linked to a MedLab biological signal collecting and processing system at day 64.Results: At day 64, pulmonary artery pressure and systemic blood pressure of the model group were higher than those of control group(P<0.05); pulmonary artery pressure and systemic blood pressure of the treatment group were lower than those of the model group(P<0.05), and systemic blood pressure was even lower than that of control group(P<0.05).Conclusion: Sildenafil could reduce pulmonary artery pressure, at the same time, also reduce systemic pressure
