Abstract:

目的:探讨大蒜素后处理对大鼠离体心肌缺血-再灌注(IR)损伤的影响。方法:24只大鼠随机分为IR组及5、10、20 μmol/L大蒜素后处理组(LA、MA、HA组),每组6只。4组应用Langendorff离体心脏灌流技术制备心肌IR损伤模型,IR组灌流稳定20 min后缺血30 min再灌注60 min; LA、MA、HA组缺血30 min后分别用5、10、20 μmol/L大蒜素灌注20 min,再用K-H液灌注40 min。记录平衡灌注末及再灌注末时的心率、左室内压上升最大速率、左室内压下降最大速率、左心室发展压和冠脉流量,检测心脏灌流液中乳酸脱氢酶(LDH)活性,左心室心肌组织中超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量。结果:与IR组比较,再灌注末时,LA、MA、HA组心功能各项指标改善,心脏灌流液中LDH活性降低,心肌组织中SOD活性提高,MDA含量降低(P<0.05)。MA组改善效果最佳(P<0.05)。结论: 大蒜素后处理可减轻大鼠离体心肌IR损伤,其适宜浓度为10 μmol/L。
Aim:To explore the effect of allicin postconditioning on myocardial ischemia-reperfusion(IR)injury in rats.Methods:A total of 24 Wistar rats were randomly allocated into 4 groups: IR group,and LA,MA,HA groups, with 6 rats in each group.Langendorff perfusion apparatus was used to build an isolated rat myocardial IR model.IR group was subjected to 30 min global ischemia followed by 60 min reperfusion after 20 min equilibrium; the LA,MA and HA groups were reperfused with 5,10,20 μmol/L allicin for 20 min,and K-H solution for 40 min after 20 min ischemia.The heart rate,left ventricular pressure maximum increase rate,left ventricular pressure drop maximum rate,left ventricular develop pressure and coronary flow were recorded at the end of 20 min equilibrium and 60 min reperfusion,the activity of lactate dehydrogenase(LDH)in heart leak,as well as the activity of superoxide dismutase(SOD)and the content of malondialdehyde(MDA)in myocardial tissue were determined.Results:Compared with IR group,cardiac function indicators improved,the activity of LDH was reduced,SOD activity increased,and MDA content was reduced in LA,MA,HA groups(P<0.05).The results of MA group were relatively better(P<0.05).Conclusion:The allicin postconditioning can reduce the myocardial IR injury, with a suitable concentration of 10 μmol/L
