Abstract:

将花生组蛋白去乙酰化酶1基因(〖WTBX〗Arachis hygogaea histone deacetylase 1, AhHDA1〖WTBZ〗)转化拟南芥野生型〖WTBX〗Col-0〖WTBZ〗和突变体〖WTBX〗had6〖WTBZ〗,对获得的纯合转基因植株进行抗旱性分析,并对ABA合成及相关响应基因表达进行检测. 结果表明: AhHDA1 蛋白定位于细胞核,在干旱条件下, 与〖WTBX〗hda6〖WTBZ〗突变体比较, 〖WTBX〗p35S〖KG-0.8mm〗WP MathAIBpHDA1/hda6〖WTBZ〗拟南芥植株的叶片相对含水量降低,气孔开度增大,干旱存活率降低,基本回复了Col的表型;体内ABA合成关键酶基因〖WTBX〗AtNCED3〖WTBZ〗和ABA信号途径重要转录因子〖WTBX〗AtABF3〖WTBZ〗基因的表达降低,但〖WTBX〗RD29A〖WTBZ〗基因表达提高. 而〖WTBX〗p35S〖KG-0.8mm〗WP MathAIBpHDA1/Col〖WTBZ〗较〖WTBX〗p35S〖KG-0.8mm〗WP MathAIBpHDA1/hda6〖WTBZ〗和Col 的抗旱性关键生理指标降低更加显著. 说明〖WTBX〗hda6〖WTBZ〗突变体表型回复确实由于外源〖WTBX〗AhHDA1〖WTBZ〗的表达引起的,推测AhHDA1影响植物抗旱性与ABA的合成与信号通路相关.
: 〖WTBX〗AhHDA1〖WTBZ〗(〖WTBX〗Arachis hygogaea histone deacetylase 1〖WTBZ〗) was transformed into Arabidopsis 〖WTBX〗Col-0 and hda6〖WTBZ〗 mutants. The drought resistance of the homozygous transgenic plants, the expression of ABA biosynthesis and drought-response genes were analyzed. The results showed as follows AhHDA1 protein was located in the nucleus; compared with 〖WTBX〗hda6 mutants,〖WTBZ〗 both the relative water content and survival rates of leaves of 〖WTBX〗p35S〖KG-0.8mm〗WP MathAIBpHDA1/hda6〖WTBZ〗 plants under drought were lower, and the stomatal apertures were higher under drought treatment, which nearly recovered Col phenotype. The expression of the key enzyme gene 〖WTBX〗AtNCED3〖WTBZ〗 in ABA biosynthesis and the key transcription factor 〖WTBX〗AtABF3〖WTBZ〗 in ABA signal pathway in the 〖WTBX〗p35S〖KG-0.8mm〗WP MathAIBpHDA1/hda6〖WTBZ〗 plant was found significantly higher than those of 〖WTBX〗hda6〖WTBZ〗, whereas the expression of 〖WTBX〗RD29A〖WTBZ〗 was lower than that of 〖WTBX〗hda6. 〖WTBZ〗The key physical parameters mediated differential drought resistance of 〖WTBX〗p35S〖KG-0.8mm〗WP MathAIBpHDA1/Col〖WTBZ〗, 〖WTBX〗p35S〖KG-0.8mm〗WP MathAIBpHDA1/hda6〖WTBZ〗 and Col. It indicated that transformed 〖WTBX〗AhHDA1〖WTBZ〗 recovered 〖WTBX〗hda6〖WTBZ〗 mutants phenotype, which might affect drought response by ABA biosynthesis and signaling
