维生素C促进根尖牙乳头间充质干细胞组蛋白去甲基化酶KDM2B和KDM5C表达的研究

Abstract:

摘要 目的:研究维生素C是否具有调节根尖牙乳头间充质干细胞组蛋白去甲基化酶相关基因表达的功能。方法:利用不同剂量的维生素C刺激人根尖牙乳头间充质干细胞;采用qRT-PCR(real time reverse transcription-polymerase chain reaction,实时定量RT-PCR)在mRNA水平检测组蛋白去甲基化酶相关基因的表达。结果:qRT-PCR结果显示组蛋白去甲基化酶KDM2B(Lysine (K)-specific demethylase 2B)和KDM5C (Lysine (K)-specific demethylase 5C)在10 μmol/L维生素C刺激后2、4和8 h表达明显升高,其表达升高程度与维生素C的作用具有时依赖性;qRT-PCR结果显示与未刺激组相比,5、10和20 μmol/L维生素C在作用8 h后都能促进 KDM2B和KDM5C的表达,其表达升高程度与维生素C的作用具有剂量依赖性的关系。结论:在根尖牙乳头间充质干细胞中维生素C可以促进组蛋白去甲基化酶KDM2B和KDM5C的表达
