巨细胞病毒宫内感染对子代豚鼠海马神经元Tau蛋白磷酸化的影响 Effect of Tau Protein Phosphorylation in the Descendant Guinea Pig with Cytomegalovirus Intrauterine Infection

Abstract:

目的:研究巨细胞病毒(CMV)宫内感染对子代豚鼠海马神经元Tau蛋白磷酸化的影响。方法:建立豚鼠CMV宫内感染动物模型,PCR法筛选CMV DNA阴性豚鼠,雌雄同笼受孕;孕龄1-20d时,随机分成两组:感染组腹腔内接种CMV病毒悬液,对照组接种生理盐水,足月分娩的子代豚鼠喂养至3月龄后无菌采集脑组织,应用免疫组织化学法和免疫印迹法检测各组子代豚鼠海马神经元Tau、p-Tau(Ser 404)、PKA及PP2A蛋白表达水平。结果:与对照组相比,感染组子代豚鼠海马神经元Tau、p-Tau(Ser 404)及PKA蛋白的表达水平明显上升,而PP2A蛋白的表达水平明显下降,差异具有统计学意义(P<0.01)。结论:CMV宫内感染导致子代豚鼠海马神经元Tau蛋白磷酸化水平增加,可能与PKA及PP2A蛋白的表达异常相关
