白术二倍体及其同源四倍体的MSAP和ISSR分析Keywords: 白术, 同源四倍体, 基因组, ISSR, MSAPAtractylodes macrocephala, autotetraploid, genome, ISSR, MSAP Abstract: [目的] 本文旨在探索白术同源四倍体基因组在分子水平上的变化规律。[方法] 以二倍体及其同源四倍体白术的试管苗为试验材料,应用MSAP和ISSR技术研究其DNA甲基化情况及遗传差异性。[结果] 白术二倍体及其同源四倍体MSAP扩增总条带数均为373条,四倍体半甲基化率高于二倍体,而总甲基化率和全甲基化率却低于二倍体,54.96%的四倍体DNA甲基化模式发生了变化。采用5条ISSR引物检测1个白术二倍体和5个同源四倍体株系,共发现25个清晰的扩增位点,多态性位点13个,多态性为52.0%。[结论] 白术二倍体人工加倍后遗传机制发生了变化。[Objectives] The aim of the paper is to explore the genomic variation at the molecular level of autotetraploid of Atractylodes macrocephala. [Methods] Diploid and autotetraploid plantlets were used in this study and DNA methylation and genetic differences between them were revealed by MSAP and ISSR techniques. [Results] The results showed that 373 loci were discovered in both diploid and autotetraploid of A.macrocephala,and the rate of hemimethylated of the autotetraploid was higher than that of diploid,but the rates of total methylated and full methylated of the autotetraploid were decreased,and DNA methylation patterns changed 54.96%. Twenty-five amplification loci and nineteen polymorphism loci were detected by five ISSR primers in one diploid strain and five autotetraploid strains of A.macrocephala,and the polymorphism was 52.0%. [Conclusions] The genetic mechanism of A.macrocephala have changed after artificial doubling
