预刺激淋巴细胞的伸瘤运动

Abstract:
目的对淋巴细胞的伸瘤现象进行描述,并探讨伸瘤的一些影响因素及伸瘤运动与淋巴细胞功能的关系。方法分离外周血淋巴细胞,用pha、il-2、a-cd3、pha+il-2预刺激后与口腔上皮癌细胞混合作用。于倒置显微镜下直接观察淋巴细胞的粘(伸)瘤情况;取混合细胞经冷丙酮固定并gimasa染色后在镜下观察淋巴细胞与瘤细胞作用情况,并于1、2、4、6h,取混合淋巴细胞于扫描电镜下观察伸瘤运动的发生、发展变化。结果与未预刺激组比较,pha组能提高粘瘤指数,il-2、a-cd3、pha+il-2均能提高淋巴细胞的粘(伸)瘤指数,以pha+il-2对粘(伸)瘤率的提高作用最为显著,电镜下发现在粘(伸)瘤运动过程中,外周血淋巴细胞的形态也发生变化。结论淋巴细胞预刺激能促进伸瘤运动的发生,伸瘤运动可能是淋巴细胞的一种杀伤方式。
