Abstract:

【目的】采用响应面法优化促孕灌注液中总黄酮的提取工艺,为促孕灌注液总黄酮的实际生产提供参考。【方法】采用水煎醇沉法制备促孕灌注液,以总黄酮得率为指标,在单因素试验的基础上,采用响应面的中心组合设计,考察液(mL)料(g)比(下同)、煎煮时间、提取次数3个因素对促孕灌注液中总黄酮得率的影响,对总黄酮的提取工艺参数进行优化分析。【结果】单因素试验结果表明,最佳浸泡时间为10 min,液料比为15∶1,煎煮时间为30 min,提取次数为3次。选择对总黄酮得率影响明显的液料比、煎煮时间、提取次数3个因素进行响应面优化分析试验,得到总黄酮最优提取条件为:液料比17.5∶1,煎煮38 min,提取3次。在最优提取工艺条件下,促孕灌注液总黄酮的理论得率为14.082 mg/g,实测得率为14.193 mg/g,二者误差较小。【结论】优化得到了促孕灌注液中总黄酮提取工艺,该工艺稳定、重复性好、总黄酮得率高。
【Objective】This study estimated the optimal conditions for total flavonoids extraction from Fertility-promoting intrauterine infusion liquid based on response surface methodology to provide theoretical foundation for its further processing and utilization.【Method】The Fertility-promoting intrauterine infusion liquid was prepared by decoction and alcohol precipitation.On the basis of single factor test,response surface methodology was applied to optimize the extraction conditions including water (mL) feed (g) ratio (the same below),boiling time,and extraction times.【Result】The optimum soaking time,water-feed ratio,boiling time,and extraction times based on single factor test were 10 min,15∶1,30 min,and 3 times,respectively.The water-feed ratio,boiling time and extraction times that had significant effects on the extraction rate of total flavonoids were selected for response surface optimization test.The optimal water-feed ratio,boiling time,and extraction times were 17.5∶1,38 min,and 3 times,respectively.The predicted extraction rate of 14.082 mg/g was very close to observed rate of 14.193 mg/g.【Conclusion】The optimized extraction process was stable,reproducible,and had high total flavonids yield from Fertility-promoting intrauterine infusion liquid
