黑麦基因组特异SCAR标记检测抗叶锈相关黑麦染色质向小麦的转移

Abstract:

利用黑麦基因组特异SCAR标记对小麦抗叶锈病近等基因系材料TcLr45和其背景材料Thatcher进行PCR扩增,在TcLr45中获得了单一条带,而在Thatcher中则扩增出2条与其大小不同的条带。进一步利用49个小麦抗叶锈近等基因系进行扩增,经5%聚丙烯酰胺凝胶电泳分析,该片段为TcLr45特有片段。经TcLr45×ThatcherF2分离群体验证,该标记与Lr45基因连锁距离较远。对该片段进行克隆测序,片段长度为650bp,与该引物在黑麦中的扩增序列有差异(643bp),进一步经BLAST2对这2个序列分析,相似性为97%,表明该片段为由黑麦向小麦转移的外源片段。100个小麦品种检测中有16个品种扩增出与TcLr45相同的条带。
