番茄果实果糖激酶基因的克隆及其表达分析

Abstract:

根据GeneBank中已登记的番茄果糖激酶基因的保守序列设计特异引物,以普通栽培型番茄辽园多丽的果实为材料提取总RNA,采用RT-PCR方法,扩增出果糖激酶基因cDNA序列的部分片段并克隆到pBS-T载体中.测序结果表明,LeFRK1-LeFRK3分别与番茄植物中已知该基因的核苷酸序列相似性高达99%以上,构建了一个番茄、马铃薯、烟草及拟南芥植物中FRK基因的系统进化树.采用半定量RT-PCR方法对番茄果实不同发育时期的表达进行检测,FRK1-FRK3在番茄果实发育的过程中均有表达,但在花后2~3d时表达量较高.
