D-柠檬烯对酒精性肝损伤大鼠铁过载调节作用

Abstract:

?目的建立酒精性肝损伤大鼠模型,探讨右旋柠檬烯(D-柠檬烯)对酒精性肝损伤大鼠铁过载的调节作用。方法70只Wistar雄性大鼠,适应性喂养7d,随机分为7组,每组10只,即对照组、模型组(酒精性肝损伤)、柠檬烯低、中、高剂量组(100、200、400mg/kg)、柠檬烯对照组(400mg/kg)、甘利欣组,给药6周后处死大鼠;苏木素-伊红(HE)染色观察肝组织形态结构;透射电镜观察肝脏超微结构变化,电感耦合等离子体质谱(ICP-MS)测大鼠肝脏内铁含量(LIC);酶联免疫吸附法测血浆铁调素(HEPC)、铁蛋白(SF)和血幼素(HJV)水平。结果HE染色结果显示,模型组大鼠肝小叶结构紊乱,肝组织呈弥漫性大泡性脂肪变性,肝小叶纤维组织增生明显,可见点灶状坏死;与模型组比较,柠檬烯组大鼠肝小叶结构紊乱程度、肝细胞脂肪变性程度和损害程度明显减轻;透射电镜结果显示,与模型组比较,柠檬烯组大鼠肝内质网、线粒体结构均有明显改善,脂滴明显减少;与对照组比较,模型组大鼠LIC、SF水平[分别为(279.14±5.87)μg/mL、(57.76±5.83)ng/mL]升高,HEPC、HJV含量[分别为(61.85±4.85)、(46.21±5.24)ng/mL]降低(P<0.05);与模型组比较,高剂量柠檬烯组大鼠LIC、SF水平[分别为(180.14±5.76)μg/mL、(35.39±3.94)ng/mL]明显降低,HEPC、HJV含量[分别为(81.36±4.8)、(67.29±5.89)ng/mL]明显升高(P<0.05)。结论D-柠檬烯对慢性酒精暴露大鼠肝脏损害所致铁过载具有一定拮抗作用。
