脑微透析与自动采血技术联用研究丹参素的药动学特性

Abstract:

通过微透析法联合自动采血技术研究丹参素在大鼠体内的药动学特性。微透析探针植入大鼠侧脑室,连续收集静脉注射丹参素后的透析液样品及血液样品,采用LC-MS/MS测定丹参素浓度,经回收率校正后,用WinNonlin药动学软件计算主要药动学参数。静脉注射丹参素后,丹参素在血液中、脑内的Ke分别为0.04,0.018min-1,t1/2为16.64,58.76min,AUC0-t为812.59,51.19min·mg·L-1,MRT为15.28,79.97min。建立的LC-MS/MS方法简便、高效、无干扰,可进行丹参素生物样品的检测;微透析技术与自动采血技术联用,较好的反映了丹参素在血液及大脑中的动力学特征,为药物药动学研究提供一个新视角。
