Abstract:

采用凝胶过滤色谱Sephadex G-50和反相高效液相色谱对胡萝卜籽中的抗氧化肽进行分离纯化.以DPPH自由基(2,2-二苯基-1-苦肼基)清除率为指标,测定多肽的抗氧化活性.采用电喷雾电离质谱法测定该多肽的相对分子质量. 研究结果表明: 经过分离纯化,得到一个具有较强抗氧化活性的多肽,在质量浓度为1 mg·mL-1时,其DPPH自由基清除活力达到(92.02±0.47)%,相对分子质量为339.3 u,由3个氨基酸残基组成.
In the present study,the antioxidant peptides from carrot seeds were purified by size exclusion chromatography Sephadex G-50 and RP-HPLC (reverse-phase high performance liquid chromatography). The antioxidant activity of each fraction was evaluated by DPPH (2,2-diphenyl-1-picrylhydrazyl) radical-scavenging effect. The molecular mass of the antioxidant peptide was identified by electrospray ionization mass spectrometry (ESI-MS). Finally,an antioxidant peptide with a molecular mass of 339.3 u was obtained. It exhibited strong DPPH radical-scavenging capability,and the scavenging rates reached to (92.02±0.47)% at the concentration of 1 mg·mL-1. The results indicated that carrot seed-derived peptides showed strong antioxidant activity
