Abstract:

球毛壳菌(Chaetomiumglobosum)是自然界中分布最广泛的真菌之一,因产生球毛壳菌素(chaetoglobosins)而被人们所熟知.然而,该真菌球毛壳菌素合成的分子机制尚不清楚.本研究利用RNAi技术证实C.globosum的一个聚酮合酶(polyketidesynthase)基因pks-1参与chaetoglobosinA合成.RNA干扰pks-1后,chaetoglobosinA的产量明显下降,同时沉默转化子的黑色素合成降低、菌丝变白,表明两种聚酮类化合物—黑色素和chaetoglobosinA,可能共有相同合成步骤.实验还发现pks-1在孢子生成过程中也起着重要的作用,pks-1基因沉默转化子失去了产孢的能力,聚酮类化合物可能通过未知过程调控孢子生成.C.globosum的pks-1基因的多重作用未有报道.本研究为C.globosum的chaetoglobosins生物合成以及次生代谢和真菌形态发生的关系提供了有用信息.
