Application of Cryo_sectioning Technique in Highly Lignified Tissues

It is normally difficult to cut nice sections and prepare good micrographes from the highly_lignified tissues by means of conventional sectioning methods. In this paper, the cryo-sectioning was introduced to overcome the difficulty and it was further proved to be a fast, simple and effective way of section preparation without passing a long process of softening, dehydration and embedding. The crucial steps are to select a proper concentration of glycerin, a suitable temperature of freezing and an appropriate way of section_flatting.
