Identification and Quantitation of Ceramides in Yeast Yarrowia lipolitica

Abstract:
Lipids in yeast Yarrowia lipolitica was separated by preparative Thin Layer Chromatography (TLC) to obtain ceramide. Mass Spectrum (MS) and tandem-MS were applied to confirm the existence of the ceramide in Y. lipolitica and to reveal its structure. Combined with High Performance Liquid Chromatography and Evaporative Light Scattering Detector (HPLC/ELSD), for the first time, the ceramide content with 24 carbons in Fatty Acid (FA) side chain was determined in Y. lipolitica.
